One hormone, one pill. Enveda announced Phase 1 results for ENV-308 on August 18, 2026. Across 88 healthy volunteers, the drug was well tolerated at every dose tested, with no serious adverse events, discontinuations, or dose interruptions, including the gastrointestinal side effects that push people off GLP-1 drugs like semaglutide.
Lac-Phe is the target. ENV-308 is built to mimic N-lactoyl-phenylalanine, a hormone the body produces in higher amounts during intense exercise and after eating. Earlier research linked it to appetite suppression, including weight loss in obese mice given the compound directly.
Leptin moved. Nothing else confirmed did. The trial's one biological readout was a reduction in circulating leptin, a hormone that signals fat stores to the brain. The effect was largest in volunteers who started with the highest leptin levels relative to their sex. Enveda's release calls this an exploratory signal, not proof the drug changes appetite or weight in people.
The market it's aimed at. Enveda plans a Phase 2 trial testing whether ENV-308 helps people keep weight off after they stop taking a GLP-1 drug, the regain problem that shows up once semaglutide or tirzepatide stops. That is a maintenance play, not a replacement for the injections.
A sprint moves 721 proteins. This pill targets one. Lac-Phe is one of the molecules that surged in a Cell Reports Medicine study on sprint intervals, where six 30-second sprints shifted 721 of 2,884 detected blood proteins immediately, against seven after 90 minutes of moderate cycling. That study noted none of those proteins sit on a standard blood panel. ENV-308 tries to isolate and reproduce one of them in pill form. Whatever the rest of that proteomic shift does for the body, this trial did not test it.
Not the same thing as exercise. Enveda's own materials state the trial did not show ENV-308 causes weight loss, builds muscle, or improves fitness. A safety trial in 88 people establishes that the pill is tolerated. It does not establish that swallowing it does what six sprints do.
